    TOOLS Cell Counting (CCK-8) kit is a ready-to-use solution without premixing of components. Cell Counting Kit-8, being nonradioactive, allows sensitive colorimetric assays for the determination of the number of viable cells in cell proliferation and cytotoxicity assays. WST-8 is reduced by dehydrogenases in cells to give a yellow colored product (formazan), which is soluble in the tissue culture medium. The amount of the formazan dye generated by the activity of dehydrogenases in cells is directly proportional to the number of living cells. The detection sensitivity of CCK-8 is higher than other tetrazolium salts such as MTT, XTT, MTS or WST-1.

The MTT assay involves the conversion of the water-soluble MTT (3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide) to an insoluble formazan.2-4 The formazan is then solubilized, and the concentration determined by optical density at 570 nm. The result is a sensitive assay with excellent linearity up to approximately 96 cells per well.

The transfected cells are induced by corresponding stimulator and lysed to determine the luciferase activity. The stimulatory-inducing effect of the regulatory elements is evaluated by luciferase activity. Renilla luciferase acts as an internal reference for correcting transfection efficiency to eliminate differences in cell number and transfection efficiency between wells. Firefly luciferase catalyzes the emission of Luciferin at 560 nm, and renilla luciferase catalyzes the emission of coelenterazine at 465 nm.

The intensity of luminescence is proportional to the amount of ATP present, and the amount of ATP is directly proportional to the number of viable cells within a certain range. Adding this reagent to lyse cell leads ATP production and the intensity of luminescence is proportional to ATP contents (number of live cells)
