EasyPrep HY-Midi Plasmid Extraction Kit
EasyPrep HY-Midi Plasmid Extraction Kit adopts a specially designed buffer system that is used in conjunction with isopropanol precipitation to achieve fast and efficient purification of high pure plasmid DNA. Plasmid DNA prepared by this Kit is suitable for a variety of routine applications including restriction enzyme digestion, sequencing, library screening, ligation and transformation, in vitro translation, and transfection of cell lines.
Recommended bacterial culture volume: 500-1500 ug plasmid with 100 ml bacterial culture for high-copy vectors; and 200-400 ug plasmid with 200 ml bacterial culture for low-copy vectors.

Fig 2. The plasmids extracted from 100 mL overnight (16 hr) E.coli (DH5α) culture (3 different samples) using EasyPrep HY Midi Plasmid Extraction Kit were analyzed by running electrophoresis with 300 ng of DNA samples per well, on a 1% TAE agarose gel prepared with TOOLS DNA View (TT-DNA01, BIOTOOLS) in 1X TAE buffer (TOOLS 50X TAE buffer, TT-50TA, BIOTOOLS) for 30 minutes (100V). DNA size was referenced with TOOLS 1kb Ladder (TMG1kb, BIOTOOLS).
Fig 3. After protocol optimization, we could increase about 66% yield of purified plasmid. In the range of 1.88~1.93 for 260/280 and 2.27~2.35 for 260/230, and also in the range of 0.89~1.11 for N/Q ratio as measured by Qubit.
