操作流程
A. Before you begin
- Spin down the vials before opening.
- Resuspend the vial mix by adding 450 μl DEPC water to the working condition. Mix by vortexing and spin down.
- If needed, make small volume aliquots of the reconstituted RNA. Store working aliquots store at -80°C for long term storage. Avoid multiple freeze/thaw cycles.
B. Prepare the spiking template during RNA extraction
- Keep the 1X RNA Spike-In Template on ice until it is unfrozen.
- For use as an RNA isolation control, add 10 μl of 1X RNA Spike-In Template to each RNA preparation during the lysate preparation step, which should include RNase inhibitors.
Important: Do not add the RNA Spike-In Template directly to the sample prior to RNA extraction, as it may be rapidly degraded. - Proceed with the remaining steps of the RNA extraction procedure according to the kit procedure without modification. Note the volume of final elution.
